The study of biological samples under vacuum requires a sample preparation to preserve both structural and chemical integrity of the living cell. However, the loss and redistribution of diffusible ions or molecules, and the possible modification of the cellular structure can produce measurement artefacts. Different samples preparations are possible (chemical fixation, cryo-fixation, ... ) and must be adapted for Transmission Electron Microscopy (TEM) and subsequent SIMS measurements. The aim of this study is to optimize the sample preparation and localize by TEM the area of interest before SIMS measurement performed on different Dynamic SIMS of the department SAM.
Biological samples from different origins (vegetals, animals) will be studied in the framework of various projects already started.
PhD in the field of Biology or Chemistry. An hands-one experience in Analytical Transmission Electron Microscopy (TEM) is mandatory
