PhD Structural Studies on Rhabdovirus Glycoproteins, France

This job proposal is a PhD position in our laboratory. The PhD will be hired in the context of an Initial Training Network (ITN) of the 7th PCRD involving 11 European research groups working on viral entry.

Our laboratory is involved in the elucidation of the structure and the molecular mechanism of working of the fusion machinery of rhabdoviruses. We have recently determined the structure of Vesicular Stomatitis Virus Glycoprotein (involved in viral entry: receptor recognition and membrane fusion), in both its pre- and post-fusion conformations (Roche et al., 2006, Science 1313:187-91 ; 2007, Science 315 : 843-8). We would like to determine the structure of other rhabdoviral glycoproteins.

The job will consist in the expression and purification of the soluble ectodomain of rhabdovirus (rabies virus and bovine ephemeral virus) glycoproteins. The purified glycoprotein will be biochemically and functionally characterized. The structure of the glycoproteins and the multimolecular complex they made will be studied by high resolution electron microscopy and by X-ray crystallography.

Proficiency in virology, protein expression and biochemistry would be an advantage.
