A Postdoctoral fellowship (2 years) is available within the framework of a Marie Curie Research Training Network (Immunanomap, MRTN_CT_2006_035946).
Research objectives: The topology, conformation and lateral mobility of the membrane proteins during this process play an important role in these processes. We would also like to investigate the changes of the cell surface distribution of MHC class I and II molecules. We aim to discover connections between the immunological function of MHC molecules and their supercluster formation on cells with modified expression levels by studying the rotational and lateral mobility of these receptor complexes using the available up-to-date spectroscopic and microscopic techniques (e.g. fluorescence correlation microscopy).
Adoptive transfer of antigen-specific T cells, recently shown therapeutic success in the treatment of cancers T cells, can be generated in vitro, bypassing in vivo constraints that often influence an effective T cell response. TCR molecules will be genetically modified and transfected back to T cells. The aim of genetic modification of TCR alpha and beta chains is to improve alpha and beta pairing and signal transduction efficacy of the constructs. Modified TCR chains are compared to non-modified TCR in vitro for their stochiometry and correct TCR pairing via FRET based flow cytometric and microscopic techniques. Quasi-realtime observation of the dynamics of molecular interactions, localization, and translocation between cellular compartments of immunocompetent molecules in immune synapse formation is of general interest within the Network (www.immunanomap.eu)
